چکیده

The low-molecular-weight stimulator of phosphofructokinase [Van Schaftingen, Hue & Hers (1980) Biochem. J. 192, 887-895] has been purified from rat liver. It was completely destroyed upon incubation with 0.01 M-HCl for 10min at 200C and fructose 6-phosphate and a reducing power equivalent in amount to the acid-labile organic phosphate were formed. It was therefore tentatively identified as fructose 2,6bisphosphate.

Increased ATP inhibition of liver phosphofructokinase from genetically diabetic mice.

Phosphofructokinase (ATP:D-fructose-6-phosphate 1-phosphotransferase, EC 2.7.1.11) was partially purified from the livers of genetically diabetic mice (C57BL/KsJ-db) and their lean littermates (C57BL/KsJ). These genetically diabetic mice have been shown to be hyperglucagonemic and to exhibit symptoms resembling those of maturity-onset diabetes in humans. Two isoenzymes of phosphofructokinase we...

Control of phosphofructokinase by fructose 2,6-bisphosphate in B-lymphocytes and B-chronic lymphocytic leukemia cells.

The levels of fructose 2,6-bisphosphate and glucose 1,6-bisphosphate and the activities of the key glycolytic enzymes have been studied in T- and B-lymphocytes, and in B-chronic lymphocytic leukemia cells (B-CLL). In both kinds of cells these two bisphosphorylated metabolites have been identified and are present at similar concentrations. Their phosphofructokinase, like that of other normal or ...

A Site of Regulation of Hepatic Gluconeogenesis by Glucagon*

The effect of glucagon on the rate of gluconeogenesis from substrates not involving pathways of mitochondrial metabolism has been investigated in isolated rat liver cells. From estimation of the rates of glucose formation, glucose phosphorylation, fructose 6-phosphate phosphorylation, fructose 1,6diphosphate dephosphorylation, and lactate and pyruvate formation, it is concluded that glucagon st...

Influence of glucagon on the metabolism of xylitol and dihydroxyacetone in the isolated perfused rat liver.

Glucagon stimulates glucose synthesis from xylitol and dihydroxyacetone in the isolated perfused rat liver. In addition to increasing the initial rate of glucose production from these substrates, the hormone also increases the efficiency of gluconeogenesis causing at least 90% of the substrate consumed by the liver to be converted to glucose. These effects of glucagon on gluconeogenesis are obs...
